Technology Summary This is a novel method of producing a vast library of bicyclic and knotted peptides for both therapeutic and imaging applications. There are several advantages to using such molecules for therapy such as improved target affinity, increased cell permeability and improved bio-stability. Although characteristics of these molecules are advantageous, difficulties in synthesis limit the ability to pull from large libraries for specified applications. As a solution, researchers at VCU have developed a system for large production of bicyclic and knotted peptides for therapeutic applications. The figure below represents the stability of the bicyclic peptides produced through this method, which compares the constructed peptide to other structures. Through this test, researchers found that this method of production enhances protease stability by testing each molecules binding affinity to streptavidin.
